چکیده

Protein-protein interactions (PPI) are involved in a myriad of cellular processes, and their deregulation can lead to many diseases. One such process is protein ubiquitination that requires an orchestrated action three key enzymes add ubiquitin moieties substrate proteins. Importantly, this reversible through deubiquitinating enzymes. Both deubiquitination require PPIs once classified be utilized identify small molecule inhibitors counteracting these reactions. Here, we study the protein-protein interaction between two OTUB1 OTUD6B report for first time both proteins directly interact with each other. We describe GFP-Trap immunoprecipitation as cell-based method analyze OTUD6B-OTUB1 context AlphaScreen (amplified luminescent proximity homogeneous assay) assay tool detect direct search PPI inhibitors.

Droplet microfluidics for high-throughput biological assays.

Droplet microfluidics offers significant advantages for performing high-throughput screens and sensitive assays. Droplets allow sample volumes to be significantly reduced, leading to concomitant reductions in cost. Manipulation and measurement at kilohertz speeds enable up to 10(8) samples to be screened in one day. Compartmentalization in droplets increases assay sensitivity by increasing the ...

High-throughput assays for promiscuous inhibitors.

High-throughput screening (HTS) searches large libraries of chemical compounds for those that can modulate the activity of a particular biological target; it is the dominant technique used in early-stage drug discovery. A key problem in HTS is the prevalence of nonspecific or 'promiscuous' inhibitors. These molecules have peculiar properties, act on unrelated targets and can dominate the result...

High-throughput assays of phagocytosis, phagosome maturation, and bacterial invasion.

Ingestion of foreign particles by macrophages and neutrophils and the fate of the vacuole that contains the ingested material are generally monitored by optical microscopy. Invasion of host cells by pathogenic bacteria and their intracellular proliferation are similarly studied by microscopy or by plating assays. These labor-intensive and time-consuming methods limit the number of assays that c...

Design and Implementation of High-Throughput Screening Assays.

HTS remains at the core of the drug discovery process, and so it is critical to design and implement HTS assays in a comprehensive fashion involving scientists from the disciplines of biology, chemistry, engineering, and informatics. This requires careful consideration of many options and variables, starting with the choice of screening strategy and ending with the discovery of lead compounds. ...
